Description

This thesis aims to develop a sol-gel manufacturing process for bioglasses, into which drugs will be integrated to combat bacterial infections or to act as anti-inflammatories after surgery, during the implantation of these bone substitutes. The objectives of this research will include studying the release kinetics of various therapeutic molecules (such as gentamicin, ibuprofen, vancomycin, and growth factors) integrated into bioglasses composed of the SiO2-Na2O-CaO-P2O5 system, depending on the glass composition. These bioglasses will be analyzed by NMR to determine their structure, which will allow us to establish a link with the drug release kinetics. Bioactivity tests will also be performed in vitro (using simulated physiological fluid) to evaluate the time required for apatite formation on the drug-enhanced bioglasses. Other parameters will need to be considered, such as the specific surface area and pore size of the doped bioglasses, as well as the composition of the medium, since these will influence the release kinetics of the therapeutic molecules. Cytotoxicity tests to assess cell viability and bacterial proliferation will allow for the study of the biocompatibility of the glasses through bone cell colonization studies and their antibacterial activity, respectively.
